The least destructive way to check for a leak would be to insert one of those fish cameras into the register (like the ones used in cars to check cylinder condition).

A lot of lake/cabin type of areas are also close to snow mobiling, skiing, ice fishing, etc.Either way, I would definitely recommend putting a small heater in the crawl space where you are worried.
