Do background checks on all 3 and make sure they all meet your rental criteria in case the couple move and the brother stays. 

Do a background check and get people in there, and you'll learn so much more in the doing than in the researching.

How do you let the applicant know that despite paying for the credit and background check them may not be choosen without making it seem like it is a personal thing?

I don't see any additional risks with having a higher volume of tenants as long as you're doing everything right (leases, background checks, etc).
