How early should I file for eviction

How early can I serve a notice to quit or file for eviction. Eg. how soon after non payment of rent (should I give them a week or 2 weeks to pay it before serving notice)?

Refer back to your state laws. I'm sure Pennsylvania has some statute that governs when you can file these notices.
